Pooling Evidence to Identify Cell Cycle-Regulated Genes

چکیده
Most of the biological studies have embraced statistical approaches to make inferences. It is common to have several independent experiments to test the same null hypothesis. The goal of research on pooling evidence is to combine the results of these tests to ask if there is evidence from the collection of studies to reject the null hypothesis. In this study, we evaluated four different pooling techniques (Fisher, Logit, Stouffer and Liptak) to combine the evidence from independent microarray experiments in order to identify cell cycleregulated genes. We were able to identify a better set of cell cycle-regulated genes using the pooling techniques based on our benchmark study on budding yeast (Saccharomyces cerevisiae). Our gene ontology study on time series data of both the budding yeast and the fission yeast (Schizosaccharomyces pombe) showed that the GO terms that are related to cell cycle are significantly enriched in the cell cycle-regulated genes identified using pooling techniques.

Anti-proliferative effect and apoptotic induction of sesquiterpene lactone parthenolide in a human breast cancer cell line
Parthenolide is a secondary metabolite, which naturally occurs in the feverfew plant and is responsible for its healing power. The potential of parthenolide in inhibition of cancer cell growth, alone or in combination with other anti-cancer therapeutics, have been studied in several laboratories. In this study, the effect of extracted parthenolide on the expression of seven pro-apoptotic genes,...

Lymphomas are solid tumors of immune system and Non-Hodgkin Lymphomas (NHL) is the most prevalent lymphomas; with wide ranges of histological and clinical features, it is so difficult to identify them. Herein, various bioinformatics tools (such as gene differential expressions, epigenetics and protein analysis) employed to find new treatment approach for NHL based on gene expression variation b...
متن کاملIdentification of cell cycle–regulated genes periodically expressed in U2OS cells and their regulation by FOXM1 and E2F transcription factors
We identify the cell cycle-regulated mRNA transcripts genome-wide in the osteosarcoma-derived U2OS cell line. This results in 2140 transcripts mapping to 1871 unique cell cycle-regulated genes that show periodic oscillations across multiple synchronous cell cycles. We identify genomic loci bound by the G2/M transcription factor FOXM1 by chromatin immunoprecipitation followed by high-throughput ...
متن کاملO-10: A Marked Animal-Vegetal Polarity in The Localization of Na+,K+-ATPase Activity and Its Down-Regulation Following Progesterone-Induced Maturation
Background: Polarized cells are key to the process of differentiation. Xenopus oocyte is a polarized cell that has complete blue-print to differentiate 3 germ layers following fertilization, as key determinant molecules (Proteins and RNAs) are asymmetrically localized. The objective of this work was to localize Na+, K+-ATPase activity along animal-vegetal axis of polarized Xenopus oocyte and fo...
متن کاملIdentification of key genes and pathways involved in vitiligo vulgaris by gene network analysis
Background and Aim: Vitiligo vulgaris is an acquired, chronic skin and hair condition characterized clinically by loss of melanin, which, if untreated, is typically progressive and irreversible. The aim of the present study was to identify potential genes involved in the pathogenesis of vitiligo. Methods: One dataset of mRNA expression in patients with vitiligo (GSE65127) were obtained from ...
